Plazomicin (ACHN 490) is a semi-synthetic aminoglycoside Antibiotic. Plazomicin acts as a substrate for Aminoglycoside acetyltransferase and Aminoglycoside phosphotransferase. Plazomicin is not modified by various common aminoglycoside-modifying enzymes. Plazomicin selectively inhibits MATE2-K. Plazomicin exhibits activity against multidrug-resistant bacteria, including carbapenem-resistant Enterobacterales.

The free form of the compound is prone to instability, it is advisable to consider the stable salt form (Plazomicin sulfate) that retains the same biological activity.

Plazomicin (ACHN 490) is a semi-synthetic aminoglycoside Antibiotic. Plazomicin acts as a substrate for Aminoglycoside acetyltransferase and Aminoglycoside phosphotransferase. Plazomicin is not modified by various common aminoglycoside-modifying enzymes. Plazomicin selectively inhibits MATE2-K. Plazomicin exhibits activity against multidrug-resistant bacteria, including carbapenem-resistant Enterobacterales[1][2].

In Vitro

Plazomicin exhibits MIC50/90 = 0.25-0.5 μg/mL/0.5-1 μg/mL Against Klebsiella, Escherichia, Enterobacter, Serratia, and Citrobacter species[1].
Plazomicin selectively inhibits MATE2-K and to a lesser extent MATE1, and OCT2, which are important transporters involved with tubular secretion[1].
Plazomicin (18 h) retains antibacterial activity against most aminoglycoside modifying enzyme-expressing E. coli BW25113 and E. coli BW25113 ΔbamB ΔtolC strains[2].
