17α,20β-Dihydroxy-4-pregnen-3-one (17,20β-P; 17,20β-DHP) acts as the maturation-inducing hormone (MIH) in salmonid fish and rainbow trout. 17α,20β-Dihydroxy-4-pregnen-3-one binds to oocyte-specific plasma membrane receptors and pertussis toxin-sensitive inhibitory G proteins in fish, inhibits Adenylate Cyclase and reduces intracellular cAMP via the membrane receptor-G protein coupling pathway, thereby initiating downstream signal transduction. 17α,20β-Dihydroxy-4-pregnen-3-one induces de novo synthesis of cyclin B, mediates the translation of cyclin B mRNA and the phosphorylation of cdc2, and promotes the production of maturation-promoting factor (MPF). 17α,20β-Dihydroxy-4-pregnen-3-one drives meiotic maturation of fish oocytes. 17α,20β-Dihydroxy-4-pregnen-3-one is applicable for development-related research.

Biological Activity
17α,20β-Dihydroxy-4-pregnen-3-one (17a,20β-DP) binds to two types of receptors with high affinity (Kd = 18 nM) and low affinity (Kd = 0.3 μM) on the oocyte membrane of rainbow trout at the late vitellogenic stage, and its signal transduction pathway involves pertussis toxin-sensitive inhibitory G protein (Gi)[1].
17α,20β-Dihydroxy-4-pregnen-3-one is the most potent steroid inducing oocyte maturation in amago salmon, and its endogenous levels increase sharply during oocyte maturation and ovulation. It inhibits adenylate cyclase activity in the oocyte membrane of rainbow trout, and this effect is partially mediated by the α subunit of inhibitory G protein (Gi)[1].
17α,20β-Dihydroxy-4-pregnen-3-one (17a,20β-DP) (100 nM-1 μM) selectively reduces pertussis toxin-catalyzed ADP-ribosylation of the 40 kDa Gi α subunit in the cell membrane of late vitellogenic oocytes in rainbow trout, indicating that it activates inhibitory G proteins[2].
17α,20β-Dihydroxy-4-pregnen-3-one induces maturation of fully grown immature goldfish oocytes via a membrane-mediated pathway, and initiates de novo synthesis of cyclin B through cytoplasmic 3'poly (A) elongation of pre-existing cyclin B mRNA[1].
17α,20β-Dihydroxy-4-pregnen-3-one induces the activation of maturation-promoting factor (MPF) in goldfish oocytes by triggering de novo synthesis of cyclin B; the newly synthesized cyclin B binds to inactive 35-kDa cdc2 kinase and promotes the phosphorylation of Thr161, thereby generating active cdc2 kinase[1].

Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.
Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.
Add each solvent one by one: 10% DMSO 90% (20% SBE-β-CD in Saline)
Solubility: ≥ 2.5 mg/mL (7.52 mM); Clear solution
This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 900 μL 20% SBE-β-CD in Saline, and mix evenly.
Preparation of 20% SBE-β-CD in Saline (4°C, storage for one week): 2 g SBE-β-CD powder is dissolved in 10 mL Saline, completely dissolve until clear.
