25N-N1-Nap
25N-N1-Nap is a blood-brain barrier-permeable, selective, β-arrestin2-biased 5-HT2A receptor (5-HT2AR) agonist. 25N-N1-Nap reduces Gq efficacy while preserving β-arrestin2 efficacy. 25N-N1-Nap does not induce hallucinogen-like behavioral responses, blocks hyperactivity induced by hallucinogens and psychostimulants, and induces rapid tolerance to hallucinogen-like responses. 25N-N1-Nap can be used for research on schizophrenia-related hyperactivity.

25N-N1-Nap is a β-arrestin-biased agonist of the 5-HT2A receptor, with reduced efficacy in the Gq pathway but preserved efficacy in the β-arrestin2 pathway, and it exhibits selectivity for the 5-HT2A receptor over the 5-HT2B and 5-HT2C receptors[1].
25N-N1-Nap (0.001 nM-0.1 mM; 60 min) induces significant internalization of the 5-HT2A receptor in the presence of co-expressed β-arrestin2, which is consistent with its β-arrestin-biased agonist profile[1].
