Nesfrotamig (YH32367; ABL105) is a bispecific activator targeting HER2 and 4-1BB. Nesfrotamig has K_d values of 0.48 nM and 3.36 nM for human HER2 and human 4-1BB, respectively. Nesfrotamig blocks tumor cell growth signals, activates HER2-dependent tumor-localized 4-1BB to maintain T cell survival, and induces NK cell-mediated antibody-dependent cellular cytotoxicity, thereby enhancing the cytotoxicity and tumor infiltration capacity of immune cells. Nesfrotamig exhibits a favorable safety profile and is suitable for research on HER2-positive and HER2-low tumors.

Nesfrotamig (ABL105) (10 mg/kg; i.p.; twice weekly; 4 weeks) induces complete tumor regression in h4-1BB KI mice bearing hHER2/MC38 colorectal tumors and establishes durable, tumor-specific memory T cell immunity that prevents tumor re-challenge^[1].

Flow cytometric analysis of 1X10⁶ Hela cells labeling ERBB2 with Nesfrotamig (HY-P991149, red). Cells were stained with the primary antibody at 1/200 dilution for an hour at 4℃. AF488-conjugated Goat Anti-Human IgG H&L (AF488) (HY-P83776) was used as the secondary antibody at 1/1,000 dilution for 30 minutes at 4℃. Human IgG1 kappa (HY-P99001, blue) was used as the isotype control, cells without incubation with primary antibody were used as the unlabeled control (black).
