Src

The Src family is the largest family of non-receptor tyrosine kinases, with ten human members: SRC, YES1, FYN, FGR, LYN, HCK, LCK and BLK, plus the related FRK and SRMS. Canonical members are N-terminally myristoylated, directing them to membranes, and share SH3, SH2 and kinase domains, with activity controlled by phosphorylation of the activating loop tyrosine and the C-terminal regulatory tyrosine targeted by CSK. Src-family kinases relay signals from growth factor, antigen, adhesion and immunoglobulin receptors to pathways controlling proliferation, adhesion and immune activation. v-Src of Rous sarcoma virus was the first oncogene-encoded tyrosine kinase discovered, and family members are drug targets in cancer, with dasatinib and bosutinib approved clinically.
