The anticancer agent 318 potently inhibits the proliferation of CYP1-expressing breast cancer cell lines MDA-MB-468 (IC50 = 0.08 μM) and TCDD-induced MCF-7 (IC50 = 0.006 μM), shows no toxicity toward non-tumorigenic MCF-10A cells (IC50 = 100 μM), and its activity depends on CYP1-mediated bioactivation[1].
The anticancer agent 318 (0-30 min) is metabolized by CYP1A1, CYP1A2 and CYP1B1 (but not CYP3A4) enzymes, producing four distinct metabolites via demethylenation, O-demethylation and hydroxylation reactions[1].
