CGS-25015
CGS-25015 is a mercapto-containing metalloprotease inhibitor that inhibits endothelin-converting enzyme (ECE). CGS-25015 inhibits ECE activity with an IC50 of 2.6 μM in porcine coronary artery smooth muscle preparations, and an IC50 of 18 μM in partially purified porcine aortic endothelial cell ECE preparations. CGS-25015 inhibits endothelin production and the resulting vascular smooth muscle contractile response.

Biological Activity
Description
In Vitro
ECE purified from BECs specifically hydrolyzes human big ET-116-38 but not human big ET-11-31 or human big ET-117-26[1].
ECE purified from PAECs cleaves human big ET-116-38 at a rate 2.7-times greater than the authentic substrate[1].

Cell Cytotoxicity Assay
Cytotoxicity assays are usually based on the assessment of cell membrane damage, which can also be indirectly detected by measuring cell viability. Detection methods include MTT assay, CKK-8 assay, LDH assay and ATP assay, etc.
