Pan-rasin-2 (compound 6A) is an orally active molecular glues that targets RAS. Pan-RAS-IN-2 inhibits pERK (IC50 in AsPC-1 cells: 0.3 nM). Pan-rasin-2 has significant inhibitory activity on cell proliferation of RAS mutant cell lines. Pan-rasin-2 can form ternary complexes with cyclophilin A (CYPA) and RAS (ON) proteins and the formation of ternary complexes can block the binding of RAF downstream of RAS, which has anti-tumor (such as colorectal cancer, pancreatic cancer) effects.

Pan-RAS-IN-2 significantly inhibits the proliferation of RAS mutant cells GP2D, PK-59, AsPC-1, PSN-1, RKN, Capan-1, SW620, and HCT116, with IC50 values of 0.9 nM, 2.3 nM, 2.1 nM, 5.3 nM, 4.5 nM, 2.5 nM, 0.2 nM, and 5.5 nM, respectively[1].
Pan-RAS-IN-2 also inhibits pERK in AsPC-1 cells, with an IC50 of 0.3 nM[1].

Pan-RAS-IN-2 (0.5-5 mg/kg; p.o.) demonstrates excellent antitumor effects in nude mice with pancreatic cancer[1].
Pan-RAS-IN-2 (1-10 mg/kg; in rats: i.v. (1 mg/kg) or p.o. (10 mg/kg); in beagles: i.v. (1 mg/kg) or p.o. (5 mg/kg)) has a long half-life in male SD rats and beagles, with T1/2 values of 5.7 h (rat, intravenous injection), 6.1 h (rat, gavage), 24.5 h (beagles, intravenous injection), and 22.4 h (beagles, gavage), respectively[1].

Add each solvent one by one: 10% DMSO 40% PEG300 5% Tween-80 45% Saline
Solubility: ≥ 2.5 mg/mL (2.99 mM); Clear solution
This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.
Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.
