Description

Isocannflavin B (Caflanone) is a flavonoid compound. Isocannflavin B exhibits anti-tumor and anti-viral activities. Isocannflavin B induces apoptosis, autophagy and cell cycle arrest in tumor cells, and inhibits cell proliferation. Isocannflavin B inhibits HCoV-OC43. Isocannflavin B can be used in research related to cancer and COVID-19[1][2][3].

In Vitro

Isocannflavin B (1-100 μM; 24 h) inhibits the proliferation of human breast cancer T47-D cells in a concentration-dependent manner, with significant growth arrest observed starting at 1 μM, and a strong inhibitory effect at 100 μM[1].
Isocannflavin B (1-100 μM; 24 h) exerts no significant effect on the proliferation of MDA-MB-231 human breast cancer cells at concentrations ranging from 1 μM to 100 μM[1].
Isocannflavin B (25 μM; 24 h) induces autophagic cytoplasmic vacuolization in human breast cancer T47-D cells without causing chromatin condensation or nuclear fragmentation[1].
Isocannflavin B (25-100 μM; 24 h) induces cell cycle arrest in human breast cancer T47-D cells, and does not induce cell apoptosis at concentrations of 25 μM or 100 μM[1].
Isocannflavin B (10-100 μM; 0-4 h) reduces the phosphorylation level of Akt in human breast cancer T47-D cells, but exerts no effect on the phosphorylation of Akt or Erk 1/2 in human breast cancer MDA-MB-231 cells[1].
Isocannflavin B potently inhibits human coronavirus OC43 in vitro, with an EC50 of 0.42 µM[2].

Cell Proliferation Assay[1]

Cell Line: T47-D human breast cancer cells
Concentration: 1 μM, 10 μM, 25 μM, 50 μM, 100 μM
Incubation Time: 24 h
Result: Caused cell growth arrest starting at 1 μM, with toxic effects evident from 25 μM.
Inhibited proliferation progressively stronger with increasing concentration.

Cell Cycle Analysis[1]

Cell Line: T47-D human breast cancer cells
Concentration: 25 μM, 100 μM
Incubation Time: 24 h
Result: Caused a cell cycle distribution shift at 25 μM, with 41% of cells in G0/G1, 11% in S, 48% in G2/M, and 2% in apoptosis.
Induced G0/G1 cell-cycle arrest at 100 μM, with 60% of cells in G0/G1, 12% in S, 28% in G2/M, and 0% in apoptosis.
Showed no induction of apoptotic cell death at either concentration.
In Vivo

Isocannflavin B enhances survival in preclinical mouse models of metastatic pancreatic cancer by promoting tumor cell apoptosis[3].
