Beta-lactamase Inhibitor

MBL-IN-1 (compound 41) is a β-Lactamase inhibitor, with the IC₅₀ ranging from 0.10 to 25.85 µM. MBL-IN-1 can be used for research of bacterial infections.

YJ196 (Compound 59) is a New Delhi Metallo-β-lactamase-1 (NDM-1) inhibitor.

N-Cbz-D-Cysteine (Cbz-D-Cys) (compound 5) is a potent β-lactamase II inhibitor with a K_i value of 20.1 µM.

Ent-Avibactam (NXL-104) sodium is a β-lactamase inhibitor with broad-spectrum antibacterial activity. When combined with Cefazidime (HY-B0593), rel-Avibactam sodium exhibits significant inhibitory effects on Enterobacteriaceae bacteria that produce Ambler A and C types of β-lactamases. The IC₅₀ value of rel-Avibactam sodium is much lower than that of commonly used β-lactamase inhibitors Clavulanic acid (HY-A0256) and Ticarcillin (HY-139805), demonstrating its high efficiency in inhibiting TEM-1 and P99 enzymes.

M104 peptide is an OXA-48 carbapenemase inhibitor and antibiotic potentiator. M104 peptide effectively blocks the binding and active site cavity of OXA-48. MBL-IN-7 is a potent metallo-β-lactamase (MBLs) inhibitor. The IC₅₀ values of MBL-IN-7 against NDM-1, IMP-1, VIM-1, VIM-2, and L1 are 11 nM, 69 nM, 29 nM, 12 nM, and 85 μM, respectively. MBL-IN-7 is an antibiotic adjuvant. MBL-IN-7 competitively inhibits enzyme activity primarily by coordinating with zinc ions in the active site of MBLs and forming hydrogen bonds and π interactions, thereby protecting carbapenem antibiotics from hydrolysis. MBL-IN-7 is applicable to research related to carbapenem-resistant Gram-negative bacterial infections.

ATM potentiators-1 is a quercetin derivative that synergistically enhances the antibacterial activity of Aztreonam (ATM, HY-B0129) against metallo-β-lactamase-producing Pseudomonas aeruginosa. ATM potentiators-1 inhibits NDM-1, OXA-10, VIM-2, KPC-2, and OXA-48, suppressing the efflux pump activity of Pseudomonas aeruginosa. FPI-1465 acts a dual inhibitor of serine-β-Lactamases and Penicillin-binding proteins (PBPs).FPI-1465 inhibits PBP2 (IC₅₀=1.0 µg/mL). FPI-1465 exhibits activity against β-lactamase CTX-M-15 and OXA-48 with K_ds of 0.011 and 5.3 µM, respectively.
