Alamethicin is a linear 20-amino acid antibiotic, which can induce voltage-gated conductance in model and cell membranes. Alamethicin exhibits antimicrobial activity against Gram-positive bacteria, but not Gram-negative bacteria. Alamethicin can form an amphipathic α-helical structure in biological membranes.

Biological Activity
A 40 mV decrease in the membrane dipole potential due to the cis-addition of 500 mM phlorizin leads to a fourfold increase in the activity of the cis-added peptide and a 20% enhancement of the
conductance of the first state of single Alamethicin channels[1].
The decrease in the dipole potential of the cis-leaflet facilitates the insertion of the positive pole, at the N-terminus of the Alamethicin dipole, into the membrane[1].
Alamethicin has an unusual primary sequence that includes non-standard amino acids and C- and N-terminal modifications[2].
When bound to lipid bilayers, Alamethicin forms amphipathic helices with polar residues aligning on one helical face. Neutron and X-ray scattering studies show that Alamethicin can form barrel-stave pores in model lipid multilayers composed of phosphatidylcholine[2].

Add each solvent one by one: 10% DMSO 40% PEG300 5% Tween-80 45% Saline

This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.
Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.
