BWC0977 (compound 13) (0.03-2 μg/mL) formic strongly inhibits the growth of multidrug-resistant Gram-negative and Gram-positive bacteria, with an MIC90 range of 0.03-2 μg/mL[1].
BWC0977 (0.015-40 μg/mL; up to 10 hours) formic exhibits time-dependent bactericidal activity against clinical isolates of E. coli, A. baumannii, K. pneumoniae, and P. aeruginosa[2].
BWC0977 (0.1 nM-1 mM; 40 min) formic potently inhibits the supercoiling activity of wild-type E. coli DNA gyrase, with an IC50 of 4 nM[4].
BWC0977 (200 μM; 8 h) formic acts via intercalation and specific interactions with ParC residues D79, M118 and R119[4].

BWC0977 (> 200 μM) formic exhibits over 5000-fold higher selectivity for bacterial type II topoisomerases than for human topoisomerase IIα and IIβ. Its IC50 against human topoisomerase IIα is 113 μM, and no detectable inhibitory effect on human topoisomerase IIβ is observed at the highest concentration of 200 μM[4].

BWC0977 (40-160 mg/kg; subcutaneous injection; every 6-24 hours; 1-4 doses) formic exhibits dose-dependent bactericidal efficacy in a neutropenic mouse thigh infection model, with an ED50 of 58 mg/kg when administered every 8 hours[3].
BWC0977 (3-450 mg/kg; intravenous injection; administered every 8-24 hours within 24 hours) formic exhibits bactericidal activity against various multidrug-resistant (MDR) pathogens in a neutropenic rat model of pulmonary infection[3].
