Tagmokitug (CHS-114; SRF-114) is a fully human IgG1 antibody targeting CCR8. Tagmokitug (CHS-114; SRF-114) is a fully human IgG1 antibody targeting CCR8. Tagmokitug (4 h) potently induces NK cell-mediated ADCC against Raji-huCCR8 cells, with an EC50 of 2 ng/mL[1].
Tagmokitug (3 h) potently induces macrophage-mediated antibody-dependent cellular phagocytosis (ADCP) against Raji-huCCR8 cells, with an EC50 of 13.5 ng/mL[1].
Tagmokitug (overnight treatment) selectively eliminates intratumoral regulatory T cells (Tregs) in co-culture systems of activated NK cells with dissociated tumor cells from primary renal cell carcinoma (RCC) and colorectal cancer (CRC), while preserving conventional CD4+ and CD8+ T cells, and activates NK cells and monocytes[1].

In Vivo

Tagmokitug (10 mg/kg; administered on days 6, 9, 13, 16 and 20 post tumor inoculation; 5 doses total) achieves 62.6% tumor growth inhibition in MC38 tumor-bearing huCCR8KI mice by depleting intratumoral regulatory T cells (Tregs) and remodeling the tumor immune microenvironment to expand the cytotoxic CD8+ T cell population[1].
