Cyclo(Gly-Pro)
Cyclo(Gly-Pro) is a cyclic dipeptide. Cyclo(Gly-Pro) exerts analgesic effects through opioid receptor-related pathways and reduces IFN-γ-induced cellular cytokine/chemokine biomarkers. Cyclo(Gly-Pro) inhibits induced nociception in mouse models, reduces carrageenan- and other inducer-induced paw edema and myeloperoxidase in inflamed paws, and increases the number of c-Fos-positive neurons in the midbrain periaqueductal gray. Cyclo(Gly-Pro) can be used in research related to pain and inflammatory diseases.

In Vivo
Cyclo(Gly-Pro) (10 μmol/kg; intraperitoneal injection; 1 h before acetic acid injection) produces a long-lasting analgesic effect in the acetic acid-induced writhing model[2].
Cyclo(Gly-Pro) (0.1-10 μmol/kg; intraperitoneal injection; 60 min after administration) produces receptor-dependent analgesic effects in the hot plate test[2].
Cyclo(Gly-Pro) (10 μmol/kg; intraperitoneal injection; single administration; 85 min after administration) increases neuronal c-Fos expression in the PAG[2].
Cyclo(Gly-Pro) (10 μmol/kg; intraperitoneal injection) attenuates carrageenan-induced mechanical hyperalgesia, paw edema, and neutrophil-associated MPO activity, and pretreatment administration also inhibits paw edema induced by PGE2, 5-hydroxytryptamine, and compound 48/80, respectively[2].
