ddCTP is a type of chain-terminating deoxynucleotide. ddCTP can be incorporated into the extension primer chain that lacks the 3'-hydroxyl group, thereby terminating primer extension, viral genome replication, and DNA synthesis. ddCTP can distinguish almost identical RNA through distinguishable extension products in primer extension inhibition experiments. ddCTP is the active metabolite of Zalcitabine (HY-17392), which can competitively inhibit HIV reverse transcriptase, terminate the synthesis of viral DNA chains, and thereby inhibit HIV replication[1][2][3].

ddCTP (0.1 mM; 20 min) enables discrimination of nearly identical RNA sequences via chain termination during poisoned primer extension reactions using total cellular RNA or purified in vitro transcripts, generating extension products of distinct sizes detectable by gel electrophoresis[1].
ddCTP, in addition to inhibiting viral reverse transcriptase, may also inhibit DNA polymerase γ in mammalian cells. This enzyme is responsible for mitochondrial DNA synthesis. Inhibition of DNA polymerase γ leads to decreased mitochondrial DNA content and impaired function[2].
