    Cell staining:
    Prepare Filipin III working solution in TBS-T. NCI-60 cancer cells were treated with 50 μg/mL Filipin III in the dark. After incubation for 1 hour at room temperature, cells were washed with TBS-T. Coverslips were then detached and mounted in a water-based mountant. Images were taken on an Olympus Provis fluorescence microscope equipped with a 40X oil objective, and quantification of cholesterol levels was accomplished using the particle size algorithm in MetaXpress image analysis software (Molecular Devices Corp., Sunnyvale CA)[2].
    Huang Y, et al assessed cholesterol enrichment on transsomes relative to TSPAN-GFP. NRK cells stably expressing TSPAN4-GFP were incubated with 2 μg/mL Filipin III at 37 °C. After 15 minutes of treatment, Z-stack images were acquired using confocal microscopy to measure filipin III intensity on migrasomes (IpM), TSPAN4-GFP intensity on migrasomes (ItM), filipin III intensity on RFs (IpR), and TSPAN4- GFP on RF. Strength(ItR)[3].
    Or as Kwiatkowska K, et al exposed fibroblasts (Fibroblast) to 25 μg/ml Filipin III and incubated in the dark for 45 minutes (20 °C), the staining results were obtained[4].

    Tissue staining:
    Filipin binds to cholesterol with high affinity; however, it exhibits rapid photobleaching and produces only modest natural fluorescence upon UV excitation, making its detection under confocal microscopy difficult[5].
    Reid PC et al used Filipin III to detect cholesterol accumulation in neurons. NPC cerebellar tissue was stained with Filipin III (125 μg/ml) for 2 h at room temperature in the dark. After incubation together, slides were washed with 0.5 M Tris without FBS and counterstained with fluorescent dyes. On day 22, cholesterol accumulation was observed in Purkinje neuronal cells. Or staining the NPC cortex with Filipin III, cholesterol accumulation in the axon hillock region of most neurons was observed on day 22[5].
    Novak A et al use Filipin III to detect cholesterol accumulation in tumors. Dissolve Filipin III in DMSO/PBS (1:4) to make a working solution of 50 μg/mL. Subsequently, the tissue sections were incubated at room temperature for 30 min. After rinsing, the slides were mounted in an aqueous fluorescent mountant for subsequent observation[6].
