Description

IBS007125 is a c-Maf inhibitor. IBS007125 does not alter the protein level or post-translational modification of c-Maf. IBS007125 inhibits some target genes of c-Maf (such as ITGB7 and CCR1). IBS007125 exerts anticancer effects on multiple myeloma expressing c-Maf. IBS007125 can be used for the research of multiple myeloma[1].

In Vitro

IBS007125 inhibits c-Maf transcriptional activity in HEK293T cells, as measured by reduced Ccl8 promoter-driven luciferase activity[1].
IBS007125 (18 h pre-incubation before LPS stimulation; 24 h LPS stimulation) selectively suppresses c-Maf-dependent cytokine (CCL8, IL-10) production without affecting c-Maf-independent TNFα production in LPS-stimulated BMDMs[1].
IBS007125 (0-20 μM; 5 days) suppresses proliferation of c-Maf-expressing RPMI-8226 multiple myeloma cells in a dose-dependent manner but does not affect proliferation of c-Maf-negative U266 multiple myeloma cells[1].
IBS007125 (10 μM; 2 days for c-Maf mRNA; 3 days for ITGB7 and CCR1 mRNA) modestly reduces c-Maf mRNA expression and significantly represses expression of the c-Maf target genes ITGB7 and CCR1 in RPMI-8226 multiple myeloma cells[1].
IBS007125 (10 μM; 2 days) does not affect c-Maf protein levels or post-translational modifications (as indicated by unaltered molecular weight) in RPMI-8226 multiple myeloma cells[1].

Cell Proliferation Assay[1]

Cell Line: c-Maf-expressing RPMI-8226 multiple myeloma cells, c-Maf-negative U266 multiple myeloma cells
Concentration: 0-20 μM
Incubation Time: 5 days
Result: Suppressed proliferation of RPMI-8226 cells by nearly 50% at 10 μM but had no effect on U266 cell proliferation.
Suppressed RPMI-8226 cell proliferation in a dose-dependent manner with increasing inhibition up to 20 μM.
Did not affect U266 cell proliferation across all tested concentrations.

Real Time qPCR[1]

Cell Line: RPMI-8226 multiple myeloma cells
Concentration: 10 μM
Incubation Time: 2 days (c-Maf mRNA); 3 days (ITGB7 and CCR1 mRNA)
Result: Modestly repressed c-Maf mRNA levels.
