Afimkibart (PF-06480605; RVT-3101) is a fully human monoclonal antibody that selectively inhibits trimeric tumor necrosis factor-like ligand 1A (TL1A). Afimkibart neutralizes active trimeric TL1A, blocks TL1A-induced signaling pathways. Afimkibart inhibits NF-κB activation and IFN-γ production. Afimkibart can be used for the research of inflammatory bowel disease.

Afimkibart selectively binds to functionally active trimeric recombinant human TL1A and does not bind to monomeric recombinant human TL1A[1].
Afimkibart potently inhibits trimeric recombinant human TL1A-induced NF-κB activation in human peripheral T lymphocytes and trimeric recombinant human TL1A/IL12/IL18-induced IFN-γ production in human PBMCs[1].

Immobilized TL1A/TNFSF15 Trimer Protein, Human can bind Afimkibart. The EC50 for this effect is 8.563 ng/mL. -
Flow cytometric analysis of 1.5X106 HT-29 cells with Afimkibart (HY-P990876, red). Cells were fixed with 4% paraformaldehyde. Then stained with the primary antibody at 1/200 dilution for an hour at 4℃. Goat Anti-Human IgG H&L (AF488) (HY-P83776) was used as the secondary antibody at 1/1,000 dilution for 30 minutes at 4℃. Human IgG1 kappa, Isotype Control (HY-P99001, blue) was used as the isotype control, cells without incubation with primary antibody were used as the unlabeled control (black).
