LC-1-40 is a selective NUDT1 PROTAC degrader with a DC₅₀ of 0.97 nM. LC-1-40 recruits NUDT1 to the CRBN E3 ubiquitin ligase complex and mediates its degradation via a proteasome-dependent pathway. LC-1-40 induces increased nucleotide damage and Apoptosis in tumors. LC-1-40 can be used in studies related to Mycn-amplified neuroblastoma.

In Vivo

LC-1-40 (30 mg/kg; i.p.; daily) selectively inhibits the growth of MYCN-amplified neuroblastoma xenografts in mice by inducing excessive oxidized nucleotide damage and tumor cell apoptosis, while exerting minimal effects on MYCN-non-amplified xenografts^[1].

Induced near-complete intratumoral NUDT1 depletion within 1 hour of a single 30 mg/kg intraperitoneal dose.
Potently impeded the growth of MYCN-amplified T409 patient-derived xenograft tumors, while exhibiting minimal growth inhibitory effect on MYCN-nonamplified T423 xenografts with comparable levels of NUDT1 degradation achieved in both tumor types.
Resulted in markedly elevated intratumoral 8-oxo-dG and cleaved-Caspase 3 staining in MYCN-amplified T409 tumors.
Showed no observable reduction in mouse body weight, indicating good tolerance.

For the following dissolution methods, please ensure to first prepare a clear stock solution using an In Vitro approach and then sequentially add co-solvents:
To ensure reliable experimental results, the clarified stock solution can be appropriately stored based on storage conditions. As for the working solution for In Vivo experiments, it is recommended to prepare freshly and use it on the same day.

Protocol 1

Add each solvent one by one: 10% DMSO 90% (20% SBE-β-CD in Saline)

Solubility: ≥ 5 mg/mL (5.92 mM); Clear solution

This protocol yields a clear solution of ≥ 5 mg/mL (saturation unknown).

Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (50.0 mg/mL) to 900 μL 20% SBE-β-CD in Saline, and mix evenly.

Add each solvent one by one: 10% DMSO 90% Corn Oil

Solubility: ≥ 5 mg/mL (5.92 mM); Clear solution

This protocol yields a clear solution of ≥ 5 mg/mL (saturation unknown). If the continuous dosing period exceeds half a month, please choose this protocol carefully.
