NA 0345 is a PKC inhibitor with IC50 values of 70 nM and 110 nM in the presence and absence of 12-O-tetradecanoyl-13-acetate, respectively. NA 0345 inhibits PKC and selectively suppresses the positive inotropic effect mediated by α1-adrenergic receptors.

Biological Activity
NA 0345 (10-100 nM) selectively inhibits the α1-adrenoceptor-mediated positive inotropic effect in isolated rabbit papillary muscles, without affecting the β-adrenoceptor-mediated positive inotropic effect[1].
NA 0345 (10-100 nM) does not affect α1-adrenoceptor-mediated IP1 accumulation in isolated rabbit ventricular slices[1].
