IBG3 is a Molecular glue degrader targeting BRD2 and BRD4, with DC50 values of 8.6 pM and 6.7 pM, respectively. IBG3 binds simultaneously to the tandem bromodomains of BRD2/BRD4, induces intramolecular conformational changes, enhances the affinity of BRD2/BRD4 for DCAF16, and promotes ubiquitination and degradation. IBG3 is applicable to the research of chronic myeloid leukemia.

IBG3 (6 h) potently degrades BRD4Tandem in KBM7 cells, with a DC50 value of 6.7 pM, which is more potent than IBG1 and dBET6[1].
IBG3 (1 nM-10 μM; 30 min) promotes the formation of the BRD4-DCAF16 ternary complex in vitro, with an EC50 of 32 nM, and exhibits stronger complex-stabilizing activity compared with IBG1[1].
IBG3 (1 pM-100 μM; 5 h) potently degrades BRD2 (DC50 = 8.6 pM) and BRD4 in HEK293 HiBiT knock-in cells, but shows no effect on BRD3, with higher efficiency than IBG1[1].
