Ro 44-3888 is an orally active and selective GP IIb-IIIa (fibrinogen receptor, integrin α_IIbβ₃) inhibitor with an IC₅₀ of 1.9 nM against the human target. Ro 44-3888 blocks the binding of fibrinogen to GP IIb-IIIa and inhibits human ADP-induced platelet aggregation. Ro 44‑3888 is formed via the metabolism of the double prodrug Ro 48‑3657 (HY-10309). Ro 44-3888 (compound 13) potently inhibits the binding of fibrinogen to purified human GP IIb-IIIa with an IC₅₀ of 1.9 nM, and effectively suppresses ADP-induced platelet aggregation in human platelet-rich plasma with an IC₅₀ of 38 nM^[1].

In Vivo

Ro 44-3888 (compound 13) is generated via in vivo metabolism after oral administration of Ro 48‑3657 (HY-10309). Its bioavailability is 26 % in rats, 25 % in dogs, and 33 % in rhesus monkeys; the corresponding elimination half-lives are 4.1 h, 11.4 h, and 5.1 h, respectively^[1].
