Endosidin-2 (ES2) is a selective inhibitor targeting the conserved exosome subunit EXO70. Endosidin-2 binds to the C-terminal domain of EXO70, inhibiting exocytosis and endosomal recycling, while promoting vacuolar trafficking in plant cells. Endosidin-2 interferes with the EXO70-mediated vesicle-to-plasma membrane anchoring process, leading to abnormal aggregation of auxin transporters (such as PIN2) in the cytoplasm and redirected to vacuolar degradation, while causing abnormal Golgi structure (such as cup-shaped or ring-shaped vesicles cisternae formation). Endosidin-2 can inhibit exocytosis in plant and mammalian cells and is mainly used to study the dynamic regulation of membrane trafficking (such as polar growth, vesicle sorting) .

Thromboxane B2 is a prostaglandin derivative that is released during anaphylaxis. Thromboxane B2 induces arterial contraction and platelet aggregation. Thromboxane B2 is a biological inactive product and circulating catabolite of thromboxane A2. Thromboxane B2 levels in peripheral venous blood and coronary sinus blood can be used as a diagnostic marker for angina. Thromboxane B2 level in serum is also associated with inhibition of COX-1 activity inhibition in platelets. Thromboxane B2 is a major product of prostaglandin endoperoxide metabolism. Thromboxane B2 produces dose-related decreases in airflow rate, tidal volume and dynamic lung compliance, whilst simultaneously increasing pulmonary airway resistance in dog model. Thromboxane B2 appears to be a naturally occurring bronchoactive metabolite in the bioconversion of arachidonic acid .

STX-721 is an orally active, irreversible, covalent EGFR exon 20 insertion (ex20ins) inhibitor that selectively targets ex20ins-mutant dynamic protein states. STX-721 potently inhibits the kinase activity of EGFR ex20ins mutants (NPG, ASV, SVD). STX-721 inhibits phosphorylation of EGFR (pEGFR Y1068) and downstream ERK (pERK Thr202/Tyr204), and suppresses proliferation of ex20ins-mutant Ba/F3 cells and human NSCLC cell lines (NCI-H2073 ASV KI, CUTO-14 ASV). STX-721 induces tumor regression in EGFR ex20ins-mutant PDX/CDX models. STX-721 can be used for the study of non-small cell lung cancer (NSCLC) harboring EGFR or HER2 ex20ins mutations .
