SIRT6-IN-2 (Compound 5) is a selective and competitive SIRT6 inhibitor (IC50: 34 μM). SIRT6-IN-2 increases acetylation of H3K9 and increases glucose uptake in cultured cells. SIRT6-IN-2 also reduces T cell proliferation. SIRT6-IN-2 has immunosuppressive and chemosensitizing effects.

SIRT6-IN-2 (200 μM) shows 100% inhibition of SIRT6 deacetylase activity, with an IC50 of 34 μM, and exhibits high selectivity for SIRT6 over SIRT1 and SIRT2[1].
SIRT6-IN-2 increases histone 3 lysine 9 (H3K9) acetylation in BxPC-3 cells to 1.9 times the level of vehicle-treated control cells[1].
SIRT6-IN-2 (100 μM; 18 h) enhances the uptake of the fluorescent glucose analog 2-NBDG in L6 cells to 1.4 times the control level[1].
