ATTO 740 NHS ester is a near-infrared fluorescent dye and a multimodal fluorescence/photoacoustic contrast agent with excellent near-infrared emission properties and extremely high photostability. In Vitro

ATTO 740 NHS ester (10 mg/mL; up to 1 h) successfully conjugates to the N-terminal amine of R01 peptide to form A740-R01, which binds integrin αᵥβ6 with a IC50 of 39.4 nM[1].

In Vivo

The A740-R01 conjugate (200 μL of 6×10-5 M; i.v.; single dose), synthesized using ATTO 740 NHS ester, enables specific detection of integrin αvβ6-positive tumors, producing a 2-fold photoacoustic signal increase and 8-fold fluorescence signal increase in positive xenografts relative to baseline 1 h after injection[1].

Animal Model: nude mice model (8-week-old female, Charles River)[1]
Dosage: 200 μL of 6×10-5 M A740-R01
Administration: i.v.; single dose
Result: Produced a 2-fold photoacoustic signal increase (1.87 arbitrary units vs. baseline 0.99 arbitrary units) in integrin αvβ6-positive A431 xenografts 1 h after injection.
Produced an 8-fold fluorescence signal increase (8.27 arbitrary units vs. baseline 1.01 arbitrary units) in integrin αvβ6-positive A431 xenografts 1 h after injection.
Showed no significant photoacoustic signal changes (pre-injection: 0.96 arbitrary units; 1 h post-injection: 1.07 arbitrary units) in integrin αvβ6-negative 293T xenografts.
Showed no significant fluorescence signal changes (pre-injection: 1.00 arbitrary units; 1 h post-injection: 1.10 arbitrary units) in integrin αvβ6-negative 293T xenografts.
Exhibited the highest maximum radiance in the liver, followed by the intestines, spleen, and kidneys in ex vivo fluorescence imaging 1 h after injection.
Detected increased photoacoustic signal in the liver at 1 h after injection, temporally dependent increases in intestinal signals peaking at 4 h, and minimal renal contrast throughout the study.
