    Folinic acid (Leucovorin) is a biological folic acid and is generally administered along with Methotrexate (HY-14519) as a rescue agent to decrease MTX-induced toxicity[1].

    In Vitro

    Methotrexate (MTX) alone induces a concentration-related increase in % micronucleated binucleated cells (MNBN) and % aberrant cells (Abs). There is a decrease in nuclear division index (NDI) with increase in MTX concentration. Similarly, the mitotic index (MI) also decreases in all concentrations of MTX tested. The addition of Folinic acid at 50 μg/ mL significantly reduces % MNBN (40-68%) and % Abs (36-77%). Inhibition is also seen at 5 μg/ mL Folinic acid (12 to 54% for MNBN and 20 to 61% for Abs) [1].

    In Vivo

    Folinic acid (7.0 mg/kg; intraperitoneal injection; every second day; for 3 weeks; Balb/c young growing male mice) treatment following Methotrexate (MTX) administration appears to reverse this growth inhibition (Chronic administration of MTX induces suppression of skeletal growth in mice)[2].

    To ensure reliable experimental results, the clarified stock solution can be appropriately stored based on storage conditions. As for the working solution for in vivo experiments, it is recommended to prepare freshly and use it on the same day.
    The percentages shown for the solvents indicate their volumetric ratio in the final prepared solution. If precipitation or phase separation occurs during preparation, heat and/or sonication can be used to aid dissolution.

    • Protocol 1

      Add each solvent one by one:  10% DMSO    40% PEG300    5% Tween-80    45% Saline

      Solubility: ≥ 2.08 mg/mL (4.39 mM); Clear solution

      This protocol yields a clear solution of ≥ 2.08 mg/mL (saturation unknown).

      Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (20.8 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.

      Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.

      Preparation of 20% SBE-β-CD in Saline (4°C, storage for one week): 2 g SBE-β-CD powder is dissolved in 10 mL Saline, completely dissolve until clear.
