Bisphenol B is a close structural analog of Bisphenol A (BPA) (HY-18260). Bisphenol B is a potent, orally active endocrine disruptor (ED). Bisphenol B binds to G protein-coupled estrogen receptor (GPER) (IC50 = 3.3 μM) with higher affinity and agonistic activity than BPA. Bisphenol B promotes GPER mediated cell migration. Bisphenol B exerts estrogenic effects via GPER pathway at nanomolar concentration. Bisphenol B is used in the manufacture of polycarbonate resin with ED properties.

Bisphenol B (Compound BPB) presents much stronger binding affinity to GPER (IC50 = 3.3 μM) than BPA (IC50 = 25.3 μM) in SKBR3 cells[3].
Bisphenol B has an EC50 of 1.7 μM for calcium mobilization, suggesting a stronger agonistic activity than BPA (EC50 = 7.5 μM)[3].
Bisphenol B (1 μM) shows higher potency in activating GPER subsequent signaling pathways than BPA in SKBR3 cells[3].
Bisphenol B (1 μM) induces SKBR3 cell migration in a concentration-dependent manner[3].
Bisphenol B (0.1 nM-10 μM) stimulates the increase of calcium mobilization and cAMP production in a concentration-dependent manner with the lowest effective concentration of 10 nM[3].
Bisphenol B (0-100 ng/mL, 2 h in CO2 incubator) significantly increases the ROS levels at 10 ng/mL and decreases the testosterone levels in testicular slices[4].

Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.
Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.
Add each solvent one by one: 10% DMSO 90% (20% SBE-β-CD in Saline)
Solubility: ≥ 2.5 mg/mL (10.32 mM); Clear solution
This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 900 μL 20% SBE-β-CD in Saline, and mix evenly.
Preparation of 20% SBE-β-CD in Saline (4°C, storage for one week): 2 g SBE-β-CD powder is dissolved in 10 mL Saline, completely dissolve until clear.
