H-Gly-Pro-Hyp-OH is a collagen-derived peptide and also a dipeptidyl peptidase-4 inhibitor.\nH-Gly-Pro-Hyp-OH inhibits the activity of dipeptidyl peptidase-4 in vitro. H-Gly-Pro-Hyp-OH binds to the DNA-binding site of the JUN/FOS complex, blocks the formation of the DNA-JUN/FOS complex, and inhibits transcriptional activity. H-Gly-Pro-Hyp-OH is applicable to research related to skin photoaging, UVB-induced skin aging/photoaging, and thrombosis[1][2][3][4].

In Vitro

H-Gly-Pro-Hyp-OH (50-100 μg/mL; 24 h) reduces UVB-induced MMP-1 production and increases type 1 procollagen synthesis in human dermal fibroblast (HDF) cells[3].

In Vivo

H-Gly-Pro-Hyp-OH (0.2 mmol/kg; p.o.; single dose) administered orally to male BALB/c mice results in transient accumulation of Gly-Pro-Hyp in plasma (Cmax 14.78 nmol/mL) and skin (Cmax 8.62 nmol/g), alongside hydrolysis to Pro-Hyp which reaches higher AUC0-t values in both tissues[1].
H-Gly-Pro-Hyp-OH (31.36 mg/kg per day; p.o.) exerts potent anti-photoaging activity in UV-irradiated Kunming mice, restoring skin elasticity, increasing hyaluronic acid and collagen content, reducing epidermal thickening, alleviating inflammation, and decreasing matrix metalloproteinase levels to a degree comparable to the high-Gly-Pro-Xaa collagen hydrolysate (HCH)[2].
H-Gly-Pro-Hyp-OH (Metabolite after 1000 mg/kg CPNS; p.o.; single dose) shows only minimal, non-significant increases in plasma concentrations in Sprague-Dawley rats, with an AUC0-t of 98.41 min∙nmol/mL over 180 minutes[3].
H-Gly-Pro-Hyp-OH (Metabolite after 300-500 mg/kg/day CPNS; p.o.; daily; 12 weeks) has no direct pharmacodynamic outcomes specific to it reported for this UVB-induced photoaging mouse model[3].
