Imzokitug
Based on 1 Customer Validation
Imzokitug (BMS-986340; CCR8.50) is a CCR8 binding agent. Imzokitug inhibits tumor growth in syngeneic tumors by blocking CCR8 and depleting tumor-resident CCR8+ Treg cells, while increasing CD4+/CD8+ T cell proliferation and upregulating PD-1. Imzokitug can be used for cancer research.

Animal Model:C57BL/6 (hCCR8 (v2) knockin expressing full-length human CCR8)[2]
-
Dosage:2 mg/kg; 10 mg/kg
-
Administration:i.p.; repeated dosing to at least 6 doses
-
Result:Led to ~38% TGI at 2 mg/kg and ~50% TGI at 10 mg/kg in the MC38 tumor model.
Tumor-resident CCR8+ Treg populations decreased, and CD4+/CD8+ T cell proliferation and PD-1 upregulation in CD4+/CD8+ T cells were observed 24 h post 6th dose.

Flow Cytometry analysis of Raji cells labelling CCR8 (red) with Imzokitug (HY-P991001). Cells were fixed with 4% paraformaldehyde and permeabilised with 90% methanol. Then cells were stained with the primary antibody at 1/200 dilution for an hour at 4℃. Goat Anti-Human IgG H&L (AF488) (HY-P83776) was used as the secondary antibody at 1/1,000 dilution for 30 minutes at 4℃. Human IgG1 kappa (HY-P99001, blue) was used as the isotype control, cells without incubation with primary antibody were used as the unlabeled control (black).
