Lolamicin

Lolamicin is an orally effective inhibitor that specifically targets the Gram-negative bacteria lipoprotein transport system LolCDE complex. It selectively inhibits the transmembrane transport of outer membrane lipoproteins by competitively binding to lipoprotein binding sites. Lolamicin destroys the integrity of the bacterial outer membrane, leading to cell death, and has both bactericidal and antibacterial activity. It has significant effects on multidrug-resistant Enterobacteriaceae pathogens (such as Escherichia coli and Klebsiella pneumoniae). Lolamicin can be used to inhibit the study of acute pneumonia, sepsis and other infections caused by Gram-negative bacteria.

In in vitro antibacterial experiments against Gram-negative bacteria (such as Escherichia coli, Klebsiella pneumoniae, and Enterobacter cloacae), Lolamicin (1-8 μg/mL; overnight culture) significantly reduces bacterial survival by inhibiting the lipoprotein transport system LolCDE[1].
In drug-resistant mutation screening experiments, Lolamicin (8×MIC; 2-48 h) induces LolC/LolE protein mutations (such as E195K, D264N) in Escherichia coli, Klebsiella pneumoniae, and Enterobacter cloacae, and the mutants are significantly less sensitive to lolamicin[1].
Lolamicin (3×MIC; 1.5 h) induces swelling of wild-type Escherichia coli and Klebsiella pneumoniae cells in confocal microscopy observation experiments, while resistant mutants (such as LolC-N265K and LolE-D264N) does not show this phenotype, indicating that it destroys the integrity of the outer membrane by interfering with lipoprotein transport[1].

Lolamicin (200 mg/kg; oral gavage; twice daily; 3 d) reduces lung tissue bacterial load and increases sepsis survival rate to more than 70% in acute pneumonia and sepsis models induced by Escherichia coli (AR0349) in male CD-1 mice[1].

- To ensure reliable experimental results, the clarified stock solution can be appropriately stored based on storage conditions. As for the working solution for In Vivo experiments, it is recommended to prepare freshly and use it on the same day.
- The percentages shown for the solvents indicate their volumetric ratio in the final prepared solution. If precipitation or phase separation occurs during preparation, heat and/or sonication can be used to aid dissolution.
Add each solvent one by one: 10% DMSO 40% PEG300 5% Tween-80 45% Saline
Solubility: ≥ 5 mg/mL (13.14 mM); Clear solution
This protocol yields a clear solution of ≥ 5 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (50.0 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.
Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.
