KB02-SLF is a nuclear FKBP12 covalent PROTAC degrader. KB02-SLF covalently modifies cysteine residues in DCAF16, forms a ternary complex with nuclear FKBP12, and mediates degradation via the CUL4-DDB1 E3 ubiquitin ligase pathway. KB02-SLF promotes polyubiquitination and proteasomal degradation of nucleus-localized FKBP12. KB02-SLF can be used in breast cancer research.

KB02-SLF (2 µM; 4-72 h) promotes sustained, concentration-dependent degradation of nuclear-localized FLAG-FKBP12_NLS in HEK293T cells via a polyubiquitination- and Cullin-RING ubiquitin ligase-dependent pathway[1].
KB02-SLF promotes the degradation of nuclear-localized FLAG-FKBP12_NLS in MDA-MB-231 cells[1].
KB02-SLF (2-10 µM; 2 h) promotes the formation of a complex between nuclear-localized FLAG-FKBP12NLS and DCAF16-containing components of the Cullin-RING ubiquitin ligase in HEK293T and MDA-MB-231 cells[1].
KB02-SLF (2 µM; 24 h) induces selective degradation of ACAT1 in HEK293T cells, but does not degrade BRD4, BRD2 or BRD3[1].
KB02-SLF (0.625-40 µM; 24 h) exhibits cytotoxicity against HEK293T cells, with an IC50 value of 14 µM[1].

- To ensure reliable experimental results, the clarified stock solution can be appropriately stored based on storage conditions. As for the working solution for In Vivo experiments, it is recommended to prepare freshly and use it on the same day.
- The percentages shown for the solvents indicate their volumetric ratio in the final prepared solution. If precipitation or phase separation occurs during preparation, heat and/or sonication can be used to aid dissolution.
Add each solvent one by one: 10% DMSO 40% PEG300 5% Tween-80 45% Saline
Solubility: 5 mg/mL (5.27 mM); Suspended solution; Need ultrasonic
This protocol yields a suspended solution of 5 mg/mL. Suspended solution can be used for oral and intraperitoneal injection.
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (50.0 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.
Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.
Add each solvent one by one: 10% DMSO 90% (20% SBE-β-CD in Saline)
Solubility: 5 mg/mL (5.27 mM); Suspended solution; Need ultrasonic
This protocol yields a suspended solution of 5 mg/mL. Suspended solution can be used for oral and intraperitoneal injection.
