NPT100-18A is an αSyn oligomerization inhibitor. NPT10018A rescues cleaved caspase-3 levels to control levels. NPT10018A attenuates mitochondrial oxidative stress probe levels in a compartment-specific manner and, at higher concentrations, increases ATP signals. NPT100-18A can be used for the study of Parkinson’s disease (PD)[1][2].

NPT100-18A (10 nM) reduces the amount of Triton-X100 insoluble αSyn aggregated conformers in iPSC-derived mDANs[1].
NPT100-18A (10 nM) specifically reduces mitochondrial ROS in iPSC-derived mDANs without affecting total intracellular ROS[1].
NPT100-18A (1 µM) significantly increases ATP signaling in iPSC-derived mDANs[1].
NPT100-18A (10 nM, 21 days) significantly reduces the proportion of cCasp3⁺ neurons in iPSC-derived mDANs and inhibits the αSyn-related early neuronal apoptosis pathway[1].

In Vivo

NPT100-18A (1-20 mg/kg, i.p., once daily for 90 days) significantly improved motor dysfunction in mThy1–wt–α-syn transgenic mice and reduced α-syn oligomers and related neurodegenerative pathology[2].

Result: 10 and 20 mg/kg significantly reduced foot slips.

Accumulation of α-synuclein in neurons and neurites was reduced.
SDS-resistant α-synuclein oligomers were reduced.
α-synuclein changed from a membrane component to a cytoplasmic component.
