Pochonin D ((+)-Pochonin D) is an inhibitor of heat shock protein 90 (Hsp90) with antiviral and anti-inflammatory activities. Description

Pochonin D ((+)-Pochonin D) is an inhibitor of heat shock protein 90 (Hsp90) with antiviral and anti-inflammatory activities. Pochonin D inhibits Hsp90, affects the homeostasis, folding and assembly processes of viral proteins, and reduces the replication capacity of viruses. Pochonin D reduces the infiltration of inflammatory cells, decreases the secretion of inflammatory cytokines such as TNF-α and IL-1β, and alleviates inflammatory responses. Pochonin D is a promising candidate for research on human rhinovirus (HRV) infection and cancer[1][2].

In Vitro

Pochonin D (0.4-10 μM; 2 days) potently inhibits HRV1B replication and CPE in Hela cells, with 90% antiviral activity at 2 μM and 10 μM (for 2 days), and reduces viral 5′NCR mRNA levels at 10 μM (48 h post-infection), while showing no cytotoxicity at tested concentrations[2].

In Vivo

Pochonin D (200 µg/kg; i.p.; twice) significantly inhibits HRV1B replication, reduces pulmonary and BALF pro-inflammatory mediator levels, decreases inflammatory cell infiltration, and ameliorates lung and tracheal pathology in HRV1B-infected mice[2].

Animal Model: BALB/c (4-5 weeks of age)[2]
Dosage: 200 µg/kg
Administration: i.p.; twice (1 hour pre-infection and 4 hours post-infection)
Result: Reduced relative HRV1B NCR gene expression in lung tissue compared to vehicle-treated infected mice.
Decreased lung levels of CCL2, CXCL1, IL-1β, TNF-α, and IL-6 to near naïve control levels.
Significantly reduced BALF levels of CXCL1, IL-1β, TNF-α, and IL-6 compared to vehicle-treated infected mice.
Significantly lowered the number (1×104 cells) and percentage of myeloid-derived suppressor cells (MDSCs) in BALF compared to vehicle-treated infected mice.
Reduced lung edema, hemorrhage, and inflammatory cell infiltration, with lower histological scores compared to vehicle-treated infected mice.
Ameliorated tracheal airway cilia damage relative to vehicle-treated infected mice.
