Akt TGF-β Receptor

Phosphatidylserine (Phospholipids) is a well-conserved anti-inflammatory and immunosuppressive signal. Phosphatidylserine is involved in membrane translocation and the activation of protein kinase C, participating in Akt signaling through its interaction with PIP3. The local exposure of Phosphatidylserine can interact with complement and other proteins, promoting microglial phagocytosis during critical periods of synaptic refinement. Phosphatidylserine can promote blood coagulation in the extracellular environment and acts as a "eat me" signal to clear out apoptotic cells. Phosphatidylserine can suppress inflammation in tissues by inducing TGF-β secretion and inhibiting immune responses .

Compound C5-IN-1 (Compound 7) is a selective allosteric inhibitor of complement component protein C5. Compound C5-IN-1 prevents C5 from being cleaved by C5 convertase, inhibits the cleavage of C5 into C5a and C5b, and thus blocks the formation of membrane attack complex (MAC). Compound C5-IN-1 has an IC50 of 0.77 μM and 5 nM in 50% human whole blood and 2% human serum to block MAC deposition induced by zymosan, respectively. Compound C5-IN-1 can be used to study diseases related to complement overactivation, such as paroxysmal nocturnal hemoglobinuria (PNH) and atypical hemolytic uremic syndrome (aHUS) .

Isatuximab is a monoclonal antibody targeting the transmembrane receptor and ectoenzyme CD38, a protein highly expressed on hematological malignant cells, including those in multiple myeloma (MM). Isatuximab has antitumor activity via multiple biological mechanisms, including antibody-dependent cellular-mediated cytotoxicity, complement-dependent cytotoxicity, antibody-dependent cellular phagocytosis, and direct induction of apoptosis without crosslinking. Isatuximab also directly inhibits CD38 ectoenzyme activity, which is implicated in many cellular functions .

Firefly luciferase mRNA-LNP is a lipid nanoparticle (LNP) containing firefly luciferase mRNA. Firefly luciferase mRNA-LNP exhibits a stable nanostructure, in which LNP plays a key role in effectively protecting and transporting mRNA to cells. Luciferase is a bioluminescent reporter gene for gene regulation and functional studies. Firefly Luciferase mRNA-LNP expresses firefly luciferase protein after entering the cells, which is often used for promoter activity detection or dual fluorescent molecular complementation experiments. Firefly luciferase mRNA-LNP is studied in research for RNA delivery, cell viability, and translation efficiency .

Cobra Venom Factor (CVF) is a selective activator targeting complement components C3, C5, and factor B in the complement system. After binding to factor B, Cobra Venom Factor is cleaved by factor D, forming a stable C3/C5 convertase resistant to regulatory proteins H and I. This continuously hydrolyzes C3 and C5, depleting serum complement while inducing neutrophil migration, vascular leakage, and increased TNF-α levels. Cobra Venom Factor can be used to deplete complement and mimic complement activation-related pathological states, and is applied in animal models of complement-mediated diseases such as acute respiratory distress syndrome (ARDS), sepsis, and shock. Cobra Venom Factor can be isolated from the venom of cobras (e.g., Naja atra, Naja melanoleuca, Naja kaouthia, etc.) .

Synonyms: N-Acetyl-L-serine
N-Acetylserine (N-Acetyl-L-serine) is a complement pathway modulator targeting activated third complement protein (C3b) and an amino-terminal residue (an N-terminal acetylation modification group). N-Acetylserine reacts with the exposed thioester group of C3b via its hydroxyl group, thereby blocking the covalent binding of glycerol to this thioester group. N-Acetylserine widely exists in soluble proteins of mammalian cells (accounting for approximately 80% of such proteins). N-Acetylserine has a blocking property that prevents direct Edman sequencing of proteins; deblocking is achievable through trifluoroacetic acid-catalyzed N→O acetyl migration followed by β-elimination. N-Acetylserine is suitable for sequencing of proteins with N-terminal acetylserine modification .
