Handling Instructions Technical Support

Ribostamycin sulfate (Vistamycin sulfate) is a broad-spectrum aminoglycoside Antibiotic with bactericidal activity against Gram-positive cocci, Gram-negative cocci, bacilli, and drug-resistant strains. Ribostamycin sulfate also acts as an inhibitor of protein disulfide isomerase (PDI), with a binding constant KD of 319 μM for bovine PDI. Ribostamycin sulfate targets bacterial 16S ribosomal RNA and the 30S ribosomal subunit, causing translational misreading and thereby inhibiting bacterial protein synthesis. Ribostamycin sulfate disrupts the integrity of bacterial cell membranes, induces membrane pore formation, and leads to bacterial death. Ribostamycin sulfate can be used in studies related to bacterial infections.

In Vitro

Ribostamycin sulfate can bind to purified bovine protein disulfide isomerase (PDI) with a dissociation constant of 319 μM; it can inhibit the chaperone activity of purified bovine PDI, but does not inhibit the autodegradation activity of purified bovine PDI[1].
Ribostamycin (0.25-64 μg/mL; 14 h) sulfate potently inhibits Haemophilus influenzae ATCC 49247 (MIC = 0.5 μM), Streptococcus pneumoniae ATCC 49619 (MIC = 7.2 μM), and three Escherichia coli strains (ATCC 4157, ATCC 12435, ATCC 10798; MIC range 0.9-7.2 μM), shows moderate activity against two other E. coli strains, and has very weak or no activity against Pseudomonas aeruginosa, Staphylococcus aureus, and Enterococcus faecalis strains in vitro[2].
Ribostamycin (0.25-64 μg/mL plus 2640 μM EDTA (HY-Y0682); 14 h) sulfate co-treatment with EDTA reduces the MIC of ribostamycin 8-fold against β-lactam-resistant Escherichia coli ATCC 35218, has no effect on its activity against E. coli ATCC 25922 or Pseudomonas aeruginosa ATCC 27853, and reduces its MIC against Staphylococcus aureus ATCC 12600 to <0.1 μM in vitro[2].
Ribostamycin (68 h) sulfate exhibits no cytotoxicity against HEK293 and MDCKII kidney cells[2].
Ribostamycin (72 h for MIC, 72 h exposure followed by 3-week subculture for MBC) sulfate shows no significant in vitro inhibitory or borreliacidal activity against 11 Borrelia burgdorferi sensu lato isolates[3].
Ribostamycin (500 μM-1 mM) sulfate forms linear oligomers that mediate the assembly of citrate-capped AuNPs (40 nm, 60 nm, 80 nm, 100 nm) into visible linear structures, with the most robust assemblies formed with 40 nm and 60 nm AuNPs[4].

In Vivo:

For the following dissolution methods, please prepare the working solution directly. It is recommended to prepare fresh solutions and use them promptly within a short period of time.
The percentages shown for the solvents indicate their volumetric ratio in the final prepared solution. If precipitation or phase separation occurs during preparation, heat and/or sonication can be used to aid dissolution.

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
