STX-100 (BG00011) is a humanized monoclonal antibody targeting αVβ6 integrin. STX-100 specifically binds αVβ6 integrin, blocks latent TGF-β activation via latency-associated peptide (LAP) interaction prevention, and reduces profibrotic responses. STX-100 can be used for the research of idiopathic pulmonary fibrosis, and cancer.

STX-100 (1 h pretreatment) exhibits minimal inhibitory effects on TGF-β1-induced morphological and nuclear changes in A549 cells[3].
STX-100 (1 h pretreatment) does not substantially alter actin cytoskeletal organization in A549 cells and does not prevent TGF-β1-associated changes in actin distribution[3].
STX-100 (1 h pretreatment) has limited inhibitory effects on TGF-β1-induced morphological and nuclear changes in MCF10A cells[3].
STX-100 (1 h pretreatment) does not reverse TGF-β1-induced changes in cell organization, cell-cell adhesion, nuclear morphology, or extracellular matrix structure in MCF10A cells[3].
STX-100 (1 h pretreatment) does not substantially alter TGF-β1-driven changes in fibronectin fibril organization in MCF10A cells[3].

Result:Showed three of five animals developed acute rejection on post-randomization days 21, 28, and 35, with sacrifice on days 26, 35, and 35 respectively.
Had two of five animals complete the 8-week course with good allograft function.
Demonstrated significantly decreased postrandomization rejection-free survival compared to placebo-treated animals.
Showed average avb6 staining intensity was lower compared to placebo-treated animals.
Exhibited significantly lower levels of LAP in tubular cytoplasm, plasma cells, and interstitial infiltrates in rejecting allografts compared to animals that rejected on standard immunosuppression alone.
Showed lower LTBP-1 in tubular cytoplasm in rejecting allografts compared to animals that rejected on standard immunosuppression alone.
Demonstrated lower active TGF-b1 in interstitial infiltrates in rejecting allografts compared to animals that rejected on standard immunosuppression alone.
Showed a mean serum total TGF-b1 increase of 31,474 pg/mL from randomization to sacrifice.

Immobilized Human Integrin alpha V beta 6 (ITGAV&ITGB6) can bind STX-100. The EC50 for this effect is 131.5 ng/mL. -
Flow cytometric analysis of 1X106 MCF-7 cells with STX-100 (HY-P990667, red). Cells were fixed with 4% paraformaldehyde. Then stained with the primary antibody at 1/200 dilution for an hour at 4℃. Alexa Fluor 488-conjugated AffiniPure Goat Anti-Human IgG H&L (AF488) (HY-P83776) was used as the secondary antibody at 1/1,000 dilution for 30 minutes at 4℃. Human IgG1 kappa (HY-P99001, blue) was used as the isotype control, cells without incubation with primary antibody were used as the unlabeled control (black).
