Garcinone B
Garcinone B is an NF-κB inhibitor. Garcinone B inhibits LPS-induced NF-κB-dependent transcriptional activation, NF-κB expression, NF-κB-mediated transcription, and IKK, prevents COX-2 transcription, and contributes to immune regulation. Garcinone B inhibits COX enzymes, inhibits prostaglandin E2 release, and contributes to anti-inflammation. Garcinone B blocks ACE2-spike binding, prevents viral entry, inhibits Mpro, and prevents polyprotein cleavage and replication. Garcinone B has antibacterial activity against cariogenic bacteria and bactericidal activity against S. mutans. Garcinone B can be used for research on COVID-19, tuberculosis, and dental caries.

In Vitro
Garcinone B inhibits cariogenic bacteria, with an MIC of 0.25 µg/mL against S. mutans, S. sobrinus, and S. mutans ATCC 25175, and an MIC of 0.5 µg/mL against L. acidophilus and L. casei[5].
Garcinone B (1-2 µg/mL; sampled at 0-24 h; plated at 48 h) exerts a bactericidal effect on S. mutans ATCC 25175, achieving bactericidal activity at 1 µg/mL and completely killing the bacteria within 1 h at 2 µg/mL[5].
Garcinone B reduces the cell surface hydrophobicity of S. mutans ATCC 25175 from 90% to 23%[5].
Garcinone B inhibits A23187-induced PGE2 release in C6 rat glioma cells and inhibits LPS-induced NF-κB transcriptional activation[4].
