Homobutein a natural chalcones (can be found in many medicinal plants, fruits, vegetables, spices and nuts), is a potent HDACs/NF-κB dual inhibitor with IC50s of 190 and 38 μM, respectively. Homobutein also a chelator of iron (II and III) cations, shows various activities, including anticancer, anti-inflammatory, antiparasite and antioxidation.

Homobutein (compound 15) (20, 24, 28, 32, 40 μM; 2 h) inhibits the viability of K562 cells[1].
Homobutein (2 h) inhibits TNFα-induced NF-κB activity in K562 cells[1].
Homobutein (1 μg/mL; 72 h) inhibits the growth of Toxoplasma gondii by 19.48%[2].
Homobutein (24 h) againsts W2 and D6 strains of P.falciparum with IC50s of 15.0 and 16.1 μM, respectively[3].

The NLRP3 inflammasome is a cytosolic innate immune signaling platform that integrates priming signals and danger-signal activation to promote caspase-1 activation, maturation of IL-1β and IL-18, and gasdermin D-mediated pyroptotic cell death.
