CRGDS
CRGDS is a cell-adhesive peptide targeting αvβ3 integrin. CRGDS competitively interferes with RGD-matrix protein adhesion and mediates integrin-dependent tumor cell uptake. CRGDS modulates endothelial viability, proliferation, capillary-like formation, and VEGF receptor inhibitory responses, whereas excessive presentation disrupts VEGF receptor-integrin synergy. CRGDS can be used in vascular disease and tumor research.

-
Cell Line:hMSC
-
Concentration:5 mM; 25 mM
-
Incubation Time:24 h (attachment); 96 h (proliferation); 10 min (UV conjugation)
-
Result:At 5 and 25 mM, cell attachment and proliferation were supported from day 1 to day 4.
The 25 mM functionalized grafts showed more hMSC attachment and proliferation than 5 mM functionalized grafts at both time points; 25 mM was selected for subsequent studies.
On day 4, the highest cell density among compared groups was on functionalized coaxial grafts; CCK absorbance was higher on functionalized coaxial grafts than on PCL grafts.

-
Animal Model:Sprague Dawley (8-9 months old, ~400 g)[1]
-
Dosage:5 or 25 mM (CRGDS); 25 mM plus 50 nM TGF-β1 (CRGDS/TGF-β1)
-
Administration:local graft conjugation (submersion 30 min, UV 15 min); implantation for 7 days
-
Result:In acellular grafts, cell density at explantation was higher than in PCL grafts, and endothelial vWF+ lining and α-SMA+ cells were present; PCL grafts showed sparse endothelial and smooth muscle cell populations.
The α-SMA+ smooth muscle layer was thicker than in PCL grafts, and graft wall thickness was greater than in PCL grafts.
ECM production was higher than in PCL grafts, with increased collagen and elastin deposition.
In grafts with TGF-β1, the α-SMA+ smooth muscle layer was thicker than in grafts without TGF-β1; the thickest smooth muscle layer was in grafts with TGF-β1 preseeded with rMSCs.
Grafts with TGF-β1 also showed increased collagen and elastin deposition versus PCL grafts, while ECM production tended to be lower than in grafts without TGF-β1.
