Mellein ((R)-Mellein) is an isocoumarin compound with antibacterial activity. Mellein inhibits the activity of HCV Protease, disrupts the cell wall of Xanthomonas sp., and acts as an insect trail pheromone. Mellein is applicable to research related to fungal infections, bacterial infections, and hepatitis C virus infections.

Biological Activity
Mellein can be produced in vitro via the enzymatic reaction of recombinant SACE5532, and can also be generated in the culture system of E. coli cells co-expressing SACE5532 and Sfp[1].
Mellein is only produced in trace amounts by Aspergillus ochraceus NRRL 3174 on yellow corn, whereas it is not detected in wheat, peanuts or soybeans[2].
Mellein exhibits broad-spectrum in vitro antimicrobial activity, with MIC values of 1.9-15.6 μg/mL against human bacterial pathogens, 7.8-15.6 μg/mL against human fungal pathogens, and 1.9-31.25 μg/mL against phytopathogenic Xanthomonas pathovars[3].
Mellein disrupts the cell wall and cellular integrity of Xanthomonas oryzae pv. oryzicola strain Y2[3].
Mellein binds to multiple Xanthomonas target proteins with binding energies ranging from -3.188 to -4.520 kcal/mol, indicating that it may inhibit bacterial protein synthesis, enhance host immunity, inactivate microbial enzymes, and suppress bacterial motility, virulence, and biofilm formation[3].
Mellein (1-100 μg/disk; 12 days) inhibits mycelial growth of the entomopathogenic fungi Beauveria bassiana and Metarhizium anisopliae[4].

This protocol yields a suspended solution of 2.5 mg/mL. Suspended solution can be used for oral and intraperitoneal injection.
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.
Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.
Add each solvent one by one: 10% DMSO 90% (20% SBE-β-CD in Saline)
Solubility: ≥ 2.5 mg/mL (14.03 mM); Clear solution
This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 900 μL 20% SBE-β-CD in Saline, and mix evenly.
Preparation of 20% SBE-β-CD in Saline (4°C, storage for one week): 2 g SBE-β-CD powder is dissolved in 10 mL Saline, completely dissolve until clear.
