Swertianolin is a xanthone glycoside. Swertianolin can be isolated from plants of the Gentianaceae family (e.g., Gentiana campestris and Swertia punicea). Swertianolin inhibits p38 phosphorylation and NF-κB activation, and reduces NO and ROS production. Swertianolin exhibits anti-Helicobacter pylori activity with an IC₅₀ of 6.1 μM and a minimum bactericidal concentration of 91.7 μM. Swertianolin modulates immune function in sepsis. Swertianolin is useful in the research of diseases such as Alzheimer's disease, hepatitis B, Helicobacter pylori infection, and sepsis.

Swertianolin (1.6-25 μg/mL; 8 days) suppresses the expression of HBeAg in HepG₂ cells with an IC₅₀ of 8.0 μg/mL^[2].
Swertianolin (12.5-200 μM; 24-72 h) significantly reduces the secretion of IL-10, NO, ROS, and arginase in MDSCs, inhibits MDSC proliferation, promotes their differentiation into dendritic cells, and improves T-cell activity by blocking the immunosuppressive effect of MDSCs^[3].
Swertianolin exhibits anti-Helicobacter pylori activity with an IC₅₀ of 6.1 μM and a minimum bactericidal concentration of 91.7 μM^[4].

In Vivo

Swertianolin (12.5 μg/kg; i.p.; 7 days) reduces the number of MDSCs in the marrow and spleen, increases CD4₊ T cells in the spleen, decreases immunosuppressive factors, reduces lung damage, and improves the survival rate of septic mice with secondary Legionella pneumophila infection by inhibiting p38 phosphorylation and NF-κB activation^[5].

Animal Model:

8-week-old C57BL/6 mice (weight 25-30 g) with sepsis induced by cecal ligation and puncture (CLP) ^[5]
Dosage:

12.5 μg/kg
Administration:

intraperitoneal injection; 7 days (from the 5th day after CLP surgery)
Result:

Reduced the proportion of MDSCs in the marrow (58.81% vs 87.89% in normal saline group) and spleen (28.60% vs 48.02% in normal saline group).
Increased CD4₊ T cells in the spleen (28.99% vs 20.44% in normal saline group).
Decreased plasma concentrations of IL-10, NO, ROS, and arginase; reduced lung damage and improved survival rate after Legionella pneumophila challenge by inhibiting p38 phosphorylation and NF-κB activation.

Stock Solution Working Solution

For the following dissolution methods, please ensure to first prepare a clear stock solution using an In Vitro approach and then sequentially add co-solvents:
To ensure reliable experimental results, the clarified stock solution can be appropriately stored based on storage conditions. As for the working solution for In Vivo experiments, it is recommended to prepare freshly and use it on the same day.
The percentages shown for the solvents indicate their volumetric ratio in the final prepared solution. If precipitation or phase separation occurs during preparation, heat and/or sonication can be used to aid dissolution.

Protocol 1

Add each solvent one by one: 10% DMSO 40% PEG300 5% Tween-80 45% Saline

Solubility: ≥ 2.5 mg/mL (5.73 mM); Clear solution

This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown).

Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.
