(R,R)-Polysphorin is a neolignan antimalarial agent that is isolated from the dried leaves and stems of Rhaphidophora decursiva. (R,R)-Polysphorin inhibits the growth of chloroquine-sensitive and chloroquine-resistant Plasmodium falciparum clones in vitro by mediating free radical damage to key cellular components via conjugated double bonds. (R,R)-Polysphorin also exhibits cytotoxicity against human oral epidermoid carcinoma cells[1].

In Vitro

(R,R)-Polysphorin (Compound 1) (42 h for anti-plasmodium assay; 72 h for cytotoxicity assay) potently inhibits the growth of chloroquine-sensitive (D6) and chloroquine-resistant (W2) Plasmodium falciparum clones, with IC50 values of 404 ng/mL and 368 ng/mL, respectively[1].
(R,R)-Polysphorin has an ED50 of 2 μg/mL against human oral epidermoid carcinoma cell line KB, with selectivity indices of 5 and 6 for D6 and W2 clones, respectively[1].
