JP-153
Based on 1 Customer Validation
JP-153 is a FAK-paxillin interaction inhibitor. JP-153 prevents the binding of the paxillin LD motif to the FAK FAT domain and disrupts the Src/FAK/paxillin focal adhesion complex, thereby reducing paxillin phosphorylation, YAP nuclear accumulation, and Akt activation. JP-153 selectively impairs mechanotaxis. JP-153 inhibits stiff matrix/TGF-β-induced myofibroblast differentiation and tumor cell invasion, and reduces tumor fibrosis and lung metastasis. JP-153 reduces Bleomycin-induced fibrosis. JP-153 inhibits VEGF-induced retinal endothelial proliferation, migration, and invasion, as well as pathological retinal neovascularization in oxygen-induced retinopathy. JP-153 attenuates tumor fibrosis and lung metastasis in KPC689 subcutaneous tumors. JP-153 can be used for research on pulmonary fibrosis, metastatic pancreatic cancer, and pathological retinal neovascularization.

Biological Activity
Description
In Vitro
JP-153 (0.5-1 µM; 24 h) reduces the number of viable primary human retinal endothelial cells at 0.5 and 1 µM under VEGF stimulation[2].
JP-153 (1 µM; 1 h) reduces VEGF-induced paxillin Y118 phosphorylation in primary human retinal endothelial cells without affecting unstimulated constitutive levels[2].
JP-153 (1 µM; 1 h pretreatment) uncouples paxillin Y118 and Akt signaling from FAK phosphorylation in VEGF-stimulated primary human retinal endothelial cells[2].
JP-153 (0.1-10 µM; 24 h) inhibits VEGF-induced migration of primary human retinal endothelial cells in the scratch wound healing assay[2].
JP-153 (0.10-0.50 µM; 24 h) inhibits VEGF-induced Transwell invasion of primary human retinal endothelial cells[2].
JP-153 (10 nM-10 µM; migration 24 h; proliferation/viability 48 h) selectively inhibits durotaxis of human lung fibroblasts without affecting chemotaxis, haptotaxis, survival, or proliferation[1].
JP-153 (100 nM; 24 h; 300 nM-1 µM after exposure) prevents stiffness-dependent myofibroblast activation but does not reverse established myofibroblast differentiation in human lung fibroblasts[1].
JP-153 blocks FAK-paxillin-YAP mechanotransduction during durotactic migration of human lung fibroblasts[1].
JP-153 inhibits durotaxis migration of primary fibrotic lung fibroblasts[1].

In Vivo:
Select the appropriate dissolution method based on your experimental animal and administration route.
- For the following dissolution methods, please ensure to first prepare a clear stock solution using an In Vitro approach and then sequentially add co-solvents:
- To ensure reliable experimental results, the clarified stock solution can be appropriately stored based on storage conditions. As for the working solution for In Vivo experiments, it is recommended to prepare freshly and use it on the same day.
- The percentages shown for the solvents indicate their volumetric ratio in the final prepared solution. If precipitation or phase separation occurs during preparation, heat and/or sonication can be used to aid dissolution.
