Lymphocyte activation gene-3 (LAG-3) is an immune checkpoint protein that belongs to the immunoglobulin superfamily. LAG-3 is mainly expressed on activated T cells, NK cells, B cells and plasmacytoid dendritic cells. LAG-3 has a high affinity for MHC class II molecules and can also bind to FGL1. Binding of LAG-3 to its ligands leads to the inhibition of T cell activation and proliferation, thereby regulating the immune response. In the tumor microenvironment, overexpression of LAG-3 is often observed, which helps tumor cells escape immune surveillance. LAG-3 is considered as an important target for cancer immunotherapy. In addition to its role in tumor immunity, LAG-3 is also involved in the pathogenesis of some autoimmune diseases and infectious diseases[1][2][3].
