Setiptiline
Based on 1 Customer Validation
Setiptiline (Org-8282) is a tetracyclic antidepressant and orally active 5-HT1ₑR/5-HT1FR agonist, with an EC50 of 171.0 nM, a Ki of 29.3 nM for human 5-HT1ₑR, and an EC50 of 64.6 nM for human 5-HT1FR. Setiptiline induces the dissociation of G proteins from the heterotrimeric Gαi1/Gβ1/Gγ2 complex and promotes the recruitment of β-arrestin2. Setiptiline is applicable to research related to migraine and schizophrenia.

In Vitro
Setiptiline (Serial dilutions; 20 min at 37°C plus 10 min at room temperature) acts as a potent full agonist at human 5-HT1ₑR (EC50 = 171.0 nM) and human 5-HT1F1R (EC50 = 64.6 nM) in a G protein dissociation BRET assay in HEK293T cells[1].
Setiptiline (Serial dilutions; 80 min at room temperature) binds to wild-type human 5-HT1ₑR with a Ki of 29.3 nM in a radioligand competition binding assay[1].
Setiptiline (10 μM; 30 min at room temperature, overnight resin binding) binds to human 5-HT1ₑR in a unique agonist-like pose, forming key interactions with the receptor's orthosteric pocket to stabilize the active-state G protein-coupled complex at 3.32 Å global resolution, as revealed by cryo-EM structural analysis[1].
Setiptiline (Serial dilutions; 20 min at 37°C plus 10 min at room temperature) has increased agonist potency, resulting in greater potency than 5-HT, at human 5-HT1ₑR with the E3116·55D mutation in a G protein dissociation BRET assay in HEK293T cells[1].

In Vivo:
Select the appropriate dissolution method based on your experimental animal and administration route.
- For the following dissolution methods, please ensure to first prepare a clear stock solution using an In Vitro approach and then sequentially add co-solvents:
- To ensure reliable experimental results, the clarified stock solution can be appropriately stored based on storage conditions. As for the working solution for In Vivo experiments, it is recommended to prepare freshly and use it on the same day.
- The percentages shown for the solvents indicate their volumetric ratio in the final prepared solution. If precipitation or phase separation occurs during preparation, heat and/or sonication can be used to aid dissolution.
