Harmalan is a tryptamine binding site stimulant and an imidazoline binding site ligand. Harmalan can be isolated from the leaves of Flindersia laevicarpa. Harmalan elevates 5-HT (HY-B1473A) levels and reduces 5-HIAA levels in the cerebral cortex. Harmalan increases spontaneous motor activity, induces clonic convulsions, produces anxiogenic effects, and elicits head weaving, hindlimb abduction, trunk curvature, piloerection, resting tremor, ataxia, forelimb abduction with increased muscle tone, and occasional alternating forepaw treading behaviors.

Harmalan exhibits low affinity for benzodiazepine binding sites in rat brain cell membranes, with an IC50 of 419 μM[1].
Harmalan binds to the 5-HT2 receptor sites on the cell membrane of the rat prefrontal cortex, with an IC50 value of 1.92 μM[1].
Harmalan exhibits high affinity for tryptamine binding sites in rat cerebral cortex cell membranes, with an IC50 of 0.038 μM[1].
Harmalan potently binds to the I-1 imidazoline binding site on rat renal cell membranes, with an IC50 of 46 nM[3].
Harmalan (40 min) potently binds to the I-2 imidazoline binding sites in rat whole brain cell membranes, with a Ki value of 148 nM[3].

Harmalan (1-40 mg/kg; i.v.) dose-dependently induces clonic convulsions in female Wistar rats, an effect antagonized by benzodiazepine agonist but not benzodiazepine site blockers, and modulated by serotonin/tryptamine receptor agents[1].
Harmalan (34 µmol/kg; i.v.) increases cerebral cortex 5-HT concentrations and decreases 5-HIAA concentrations in female Wistar rats, with additive effects when combined with monoamine oxidase inhibitor and indolealkylamine[1].

Result:Caused lateral head weaving, hind limb abduction, back bending, coat ruffling, and resting torso tremor in 8/10 rats at 3.125 mg/kg.
Induced head and torso resting tremor lasting 2-3 hours, ataxia, forelimb abduction, increased extensor muscle tone, and occasional reciprocal forepaw treading in all rats, plus clonic convulsions followed by postictal relaxation in 3/10 rats at 6.25 mg/kg.
Triggered seizure trains lasting up to 15 minutes post-injection in 7/10 rats at 9.375 mg/kg.
Produced clonic convulsions in 9/10 rats at 10 mg/kg.
Induced marked excitation with generalized convulsions and jumping up to 0.5 m in all rats at 12.5 mg/kg.
