A-75943
A-75943 is a bone resorption inhibitor isolated from Streptomyces sp. SANK 61296, with protein synthesis toxicity. A-75943 inhibits bone resorption in vitro, and also suppresses the increase in serum calcium levels and the decrease in bone mineral density in thyroparathyroidectomized rats. A-75943 inhibits protein synthesis. High concentrations of A-75943 reduce the survival rate of osteoclasts on bone slices, while low concentrations have no effect on the survival rate or morphology of osteoclasts. A-75943 can be used for the research of osteoporosis and hypercalcemia.

A-75943 (5 μg/mL; 2 days) inhibits bone resorption in mouse unfractionated bone cell cultures with an IC50 of 0.35 μM, achieving >90% inhibition at 5 μg/mL without cytotoxicity at lower concentrations[1].
A-75943 inhibits protein synthesis in vitro with an IC50 of 153 μM[1].
