Aurachin D is an antibiotic and selective cytochrome bd oxidase inhibitor. Aurachin D exhibits inhibitory effects against Gram-positive bacteria and a small number of fungi, with an IC50 value of 0.1 μM against Escherichia coli and an IC50 value ranging from 0.13 to 33 μM against Mycobacterium tuberculosis. Aurachin D binds to and inhibits cytochrome bd oxidase, thereby blocking the electron transfer process, disrupting the respiratory chain and energy metabolism of pathogens. Aurachin D can be used in research related to leishmaniasis, malaria, Chagas disease, African trypanosomiasis, and tuberculosis[2].

Aurachin D selectively inhibits Escherichia coli cytochrome bd-II oxidase by binding to the hydrophobic pocket in the AppC subunit, stabilizing the Q-loop, and blocking quinol oxidation at the enzyme active site[2].
Aurachin D (50 nM; 4 min) inhibits NADH oxidation in bovine heart submitochondrial particles, with an IC50 of 50 nM[1].
Aurachin D potently inhibits the sodium-translocating NADH:ubiquinone oxidoreductase (Na+-NQR) of Vibrio cholerae by binding to the cytoplasmic surface of its NqrB subunit, which partially overlaps with the ubiquinol-binding site[2].
Aurachin D (up to 512 μM, 48 h-7 days) potently inhibits bacterial growth in M. tuberculosis mc26230 (MIC = 8 μM), but shows no obvious antibacterial activity in M. smegmatis mc2155 Wt and the Δqcr mutant (MIC > 512 μM)[4].\n
Aurachin D potently inhibits cytochrome bd oxidase in inverted membrane vesicles (IMVs) containing M. tuberculosis cytochrome bd oxidase and reduces oxygen consumption (IC50 = 0.15 μM)[4].
Aurachin D (48-72 h) exhibits potent anti-parasitic activity against Plasmodium falciparum NF54 (erythrocytic stage), Trypanosoma brucei rhodesiense STIB 900 (bloodstream form), Trypanosoma cruzi Tulahuen C4 (intracellular amastigote) and Leishmania donovani MHOM-ET-67/L82 (axenic amastigote), with IC50 values of 0.012 μM, 4.5 μM, 1.3 μM and 0.044 μM, respectively. It shows low cytotoxicity against rat skeletal muscle myoblast L6 cells (IC50 = 130.7 μM)[5].
