MC4491
MC4491 is a selective LSD1/PRMT5 inhibitor, with an IC50 of 0.152 μM against the LSD1/CoREST complex and an IC50 of 0.043 μM against the PRMT5/MEP50 complex. MC4491 induces transcriptomic changes and splicing alterations in AML cells. MC4491 is applicable for the research of acute myeloid leukemia.

MC4491 potently and selectively inhibits both LSD1/CoREST (IC50 = 0.152 μM) and PRMT5/MEP50 (IC50 = 0.043 nM) in biochemical assays, with minimal activity against PRMT1 and PRMT7[1].
MC4491 (50 nM; 3 days) treatment of MV4-11 AML cells recapitulates the transcriptomic and alternative splicing changes induced by combined PRMT5 and LSD1 inhibition, including activation of myeloid differentiation pathways and suppression of cell cycle-related pathways[1].
