MC4491 potently and selectively inhibits both LSD1/CoREST (IC₅₀ = 0.152 μM) and PRMT5/MEP50 (IC₅₀ = 0.043 nM) in biochemical assays, with minimal activity against PRMT1 and PRMT7^[1].
MC4491 (50 nM; 3 days) treatment of MV4-11 AML cells recapitulates the transcriptomic and alternative splicing changes induced by combined PRMT5 and LSD1 inhibition, including activation of myeloid differentiation pathways and suppression of cell cycle-related pathways^[1].
